Structural elucidation of one or more minor components of a mixture
by NMR is often best handled through an off-line separation of
the component into an NMR tube, followed by NMR analysis. This
process enables the often long term NMR experiments that provide
detailed structural elucidation information to occur while the
off-line LC and LC-MS apparatus is used for other separations.
However, for natural product analyses or metabolite identification
where NMR data is required for unequivocal identification, there
are a number of situations where an in-line system has benefit. For
example, some metabolites may undergo rearrangement when stored
on columns or cartridges. Or, a sample may contain two or
more natural products with very similar LC-MS or LC-MS/MS characteristics. For
either of these applications, running the output of the LC directly
into an NMR flow cell can provide optimal information.
The Varian LC-NMR and LC-NMR/MS systems provide integrated platforms
that run all the components of the system from one software - VnmrJ. This

LC-NMR/MS Elements
The ProStar HPLC Chromatography System
is used for the separation of chemical compound mixtures. Its modular concept makes it ideally suitable
for LC-NMR. Typically consisting of a ProStar 210
solvent pump and a ProStar 335 Diode Array Detector,
the ProStar HPLC is flexible enough to solve demanding separation problems.
Both pump and detector are controlled from the VnmrJ Software when used in conjunction with NMR. The HPLC
can, however, also be operated independently from the NMR machine for off-line separation of compounds using its own
PC and software.

The 320-MS Triple-Quadrupole Mass Spectrometer
is a high-sensitivity but low mainenance mass spectrometer. Its ionization sources can easily be swapped
between either Electron Spray (ESI) or Atmospheric Pressure Chemical Ionization (APCI) units to adapt to the
varying requirements of different compounds. The 320-MS can be operated either from the VnmrJ software for
LC-NMR-MS work or also independently from its own PC and software.
